UltraPlex™ Multiplexed Assays
The UltraPlex™ system has already been used to multiplex a wide range of assays which demonstrate its broad applicability. The most advanced applications are in direct immunoassay, where Pronostics has already developed and is launching a portfolio of products directed at clinical immunology laboratories. These products clearly demonstrate that UltraPlex™ assays yield sensitivities and specificities at least as good as conventional “one analyte at a time” ELISAs.
UltraPlex™ ANA is available today and provides valuable time and cost savings compared to existing ENA/ANA experiments using Hep-2 screens and ELISA tests. It allows hospitals and laboratories to perform fully automated profiles of patient samples and gives qualitative results for the following antibodies in human serum: Jo-1, SSA, SSB, Sm, Sm/RNP, U1snRNP, Scl-70 and Centromere B, and quantitative results for dsDNA. This product is fully CE marked and is initially available in the UK and other European countries.
A family of related UltraPlex™ immunoassay kits will be available during 2007 including Thyroid, Coeliac Disease and Rheumatoid assays.
UltraPlex™ FingerPrint Profiling Assays
Molecular fingerprinting is a new approach to medical diagnostics. Fingerprints based on genes or proteins, and generated by expensive equipment such as NMR machines or mass spectrometers, have shown tremendous promise in research laboratories. To date no practical and economic method has previously been devised for generating such profiles. However, UltraPlex™ now makes delivery of revolutionary profiling diagnostics, at an acceptable cost to users, a reality.
Pronostics’ is using its proprietary UltraPlex™ platform technology, antigen libraries, and pattern recognition software to create biological fingerprints focusing on profiling the complete immune system of an individual from a single simple blood sample. By measuring the levels of thousands of chemicals in a blood sample simultaneously it is possible to construct a profile of an individual which can be used to safely and accurately diagnose a wide range of different diseases. Pronostics first FingerPrint product, CADPrint™ will be commercially available during 2008.
UltraPlex™ Automation
Full “walk away”automation for UltraPlex™ solutions is provided by the UltraPlex™ SmartReader, running SmartDecode software, and the UltraPlex™ SmartStation liquid handling robot, which virtually eliminate most of the usual assay “hands on” time.
APPLICATIONS
Together with its partners for technology and application development, Pronostics is busy establishing UltraPlex™ as the preferred technology platform for scalable multiplexed testing in medical diagnostics and drug discovery.
In the IVD market place, Pronostics is currently developing a portfolio of immunoassay solutions for the autoimmune and allergy sectors and in the cardiovascular sector, its first biological fingerprinting product, CADPrint™ is at an advanced stage of development.
Applications are not limited to immunoassays. We have demonstrated that biotinylated oligonucleotides can be readily coupled to streptavidin-coated UltraCodes™, and then be specifically hybridized with labelled probes, opening up a wide range of potential applications in multiplex genetic testing.
The company has full ISO 13485:2003 accreditation and all IVD kits launched will be CE marked or carry FDA approval as appropriate.
Together with partners already established in the drug development markets, Pronostics is adding value by providing the flexibility, customisation and high-throughput multiplexing capabilities of the UltraPlex™ platform to significantly reduces costs and time scales in the discovery, development and clinical areas.
